Target description
Astrocytic phosphoprotein PEA-15 (UniProt: Q15121; also known as 15 kDa phosphoprotein enriched in astrocytes, Phosphoprotein enriched in diabetes, PED) is encoded by the PEA15 gene (Gene ID: 8682) in human.PEA-15 is a cytoplasmic protein that is ubiquitously expressed. Higher expressions are found in heart, brain, muscle, and adipose tissue. Higher expression oF PEA-15 has been reported in tissues from individuals with type 2 diabetes. PEA-15 blocks Ras-mediated inhibition of integrin activation and modulates the ERK MAP kinase cascade. It is reported to Inhibit RPS6KA3 activities by retaining it in the cytoplasm. It is also shown to inhibits both TNFRSF6- and TNFRSF1A-mediated caspase 8 activity and apoptosis. It also regulates glucose transport by controlling both the content of SLC2A1 glucose transporters on the plasma membrane and the insulin-dependent trafficking of SLC2A4 from the cell interior to the surface. PEA-15 is phosphorylated by protein kinase C and calcium-calmodulin-dependent protein kinase. These phosphorylation events are modulated by neurotransmitters or hormones. PEA-15 has two microtubule binding sites (aa 98-107 and 122-129). SMEK (suppressor of mek1) is a regulatory subunit of protein phosphatase 4 (PP4) complex. It consists of SMEK1 and SMEK2 subunits. SMEK is involved in histone deacetylation and silencing of the Wnt-signaling gene, brachyury, in embryonic stem cells (ESCs). It plays an important role in the recruitment of PP4 complex and histone deacetylase 1 (HDAC1) to the Tcf/Lef binding site of the brachyury promoter, inhibiting brachyury expression in ESCs. It associates with PP4c and represses transcription of Wnt-responsive development-related genes through histone deacetylation, maintaining the pluripotency of ESCs.

Prenylated Rab acceptor 1 domain family, member 2 (PRAF2) is a novel 19kDa protein with a prenylated Rab acceptor 1 (PRA1) motif and four transmembrane domains and belongs to the PRAF protein family. PRAME (preferentially expressed antigen in melanoma) is overexpressed in malignant cells, including primary and metastatic melanomas, acute and chronic leukaemias, Hodgkins lymphoma, breast cancer and head and neck squamous cell carcinomas. In AML (acute myeloid leukemia), high levels of PRAME are associated with lower relapse rate and high chances of disease-free survival. PRAME also plays a role in the retinol pathway. It modulates the metabolism of all-trans retinol (vitamin A) and its active metabolites, referred to as retinoids. It is a repressor of retinoic acid receptor signaling.

In renal cell carcinoma, chromosomal translocation results in PRCC (papillary renal cell carcinoma translocation-associated gene)-TFE3 (bHLH-LZ transcription factor) fusion protein. The fusion protein induces expression of cyclin-dependent kinase inhibitor p21-CIP1 (CDK-interacting protein 1) and thereby causes cell cycle delay. In addition, PRCC-TFE3 is unable to bind with PRCC-interacting mitotic checkpoint protein (MAD2B), resulting in mitotic checkpoint defect.

PRDX6 (Peroxiredoxin 6) possesses two types of activities, peroxidase and phospholipase A2 activities. It acts as a bifunctional enzyme in the cell controlling during oxidative stress. PRDX6 converts hydrogen peroxide (H2O2) and alkyl hydroperoxides to water and alcohol through redox reactions. It also functions as an antioxidant enzyme in the antioxidant defense mechanism and lung phospholipid metabolism. It has been reported that overexpressed PRDX6 reduces the chances of oxidative damage, whereas knockdown expression leads to the oxidative stress and apoptosis. Biochem/physiol Actions
The gene PREB (prolactin regulatory element binding) encodes a type II transmembrane protein. It binds to the Pit1-binding element of the prolactin (PRL) promoter and plays a transcriptional regulatory role in the pituitary and other organs in which the protein is expressed. In response to cytokines, PREB transcriptionally regulates MCP-1 (monocyte chemoattractant protein-1) promoter. MCP-1 functions as an initiating factor in the inflammatory process of atherogenesis. PREB also functions as a guanine-nucleotide exchange factor for Sar1 guanosine triphosphatase and is essential for collagen VII secretion. It is recruited to ER (endoplasmic reticulum) exit sites by its interaction with a collagen cargo receptor component, cTAGE5. The export of large cargo at the ER exit sites involves high levels of guanosine triphosphate-bound Sar1 generated by PREB.

PREX2 (phosphatidylinositol-3,4,5-trisphosphate-dependent Rac exchange factor 2) interacts with and suppresses PTEN (phosphatase and tensin homolog) protein in breast cancer. Mutations in this gene are linked to human melanomas, and abnormal expression of this gene leads to tumorigenesis in immortal melanocytes. Mutations in this gene are also associated with lung, colon and pancreatic cancer. Thus, mutations are thought to turn PREX2 in an oncogene. It recognizes and interacts with Rac-1 protein, RhoA and Cdc42, and activates them. This, in turn, is activated by PI-3K (Phosphoinositide 3-kinase). It is induced by CXCL9 (Chemokine (C-X-C Motif) Ligand 9), which results in enhanced invasiveness of hepatocellular carcinoma. PREX2 is also up-regulated in gastric cancer (GC), and this expression is suppressed by miR-338-3p, which acts as a tumor suppressor in GC.

Biochem/physiol Actions
Prickle homolog 1 is a nuclear receptor encoded by the PRICKLE1 gene. The protein is involved in the planar cell polarity pathway. This pathway controls convergent extension during gastrulation and neural tube closure. This gene may have a role in human neural tube defects. Defects in this gene have been associated with progressive myoclonus epilepsy characterized by myoclonic seizures, generalized convulsive seizures, neurologic decline such as ataxia.

Target description
Major prion protein (UniProt: P04156; also known as PrP, ASCR, PrP27-30, PrP33-35C, CD230) is encoded by the PRNP (also known as ALTPRP, PRIP, PRP) gene (Gene ID: 5621) in human. PrP is highly expressed in the brain, lung, kidney and heart and low levels of expression are reported in the liver and spleen. It is found in high quantity in the brain of humans and animals infected with degenerative neurological diseases such as Kuru, Creutzfeldt-Jakob disease (CJD), Gerstmann-Straussler syndrome (GSS), scrapie, bovine spongiform encephalopathy (BSE), and transmissible mink encephalopathy (TME). In normal brain it may play a role in neuronal development and synaptic plasticity and may be required for neuronal myelin sheath maintenance and myelin homeostasis through acting as an agonist for ADGRG6 receptor. PrP can exist in either PrPc (c = cellular) or PrPsc (sc = Scrapie) form. PrPc is a glycoprotein normally found inserted in the plasma membrane by a glycosyl phosphatidylinositol (GPI) anchor and can be easily digested by proteases. It is composed mainly of -helices. The PrPsc has more of a -sheet type structure and is highly resistant to the action of proteases. The prion proteins possess two N-linked glycosylation sites and may contain over 50 different sugars. The large size of the N-linked sugars enables them to shield two orthogonal faces of the protein almost completely and protect large regions of the protein surface from proteases. The PrPsc is shown to contain the same set of glycans as PrPc but has a higher proportion of tri- and tetra-antennary sugars. It is shown that following proteinase K treatment, PrPc is completely digested, whereas PrPsc is only shortened to a 27-30 kDa fragment. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
