Synphilin-1 contains several protein-protein interaction domains, including six ankyrin-like repeats, coiled-coil domain, and an ATP/GTP binding domain. The synphilin gene produces at least nine transcript variants encoding seven distinct proteins. Synphilin transcripts are widely expressed in many tissues with highest levels found in the brain, heart and placenta. Synphilin-1 is highly enriched in presynaptic nerve terminals.Immunogensynthetic peptide corresponding to amino acids 829-847 of human synphilin, conjugated to KLH. This sequence is identical in dog synphilin-1 and highly conserved (84% identity) in mouse, rat, and bovine synphilin-1.ApplicationAnti-Synphilin-1 antibody produced in rabbit has been used in co-immunoprecipitation and western blotting.
Biochem/physiol Actions
Synphilin-1 associates with and is ubiquitinated by several proteins, including α -synuclein, parkin, dorfin and siah E3 ubiquitin protein ligase 1 (SIAH1), and is a major component of Lewy bodies in Parkinson′s disease (PD). It is associated with synaptic vesicles and is modulated by α-synuclein. The central domain of synphilin-1 has been suggested to be required for the formation of aggregates and cytotoxicity. Synphilin-1 association with α -synuclein promotes the formation of cytosolic inclusions. Mutation of synphilin-1 in PD patients at amino acid R621C, is implicated in these sequestration, ubiquitination and proteasomal inhibition leading to increased accumulation of toxic intermediates. Cells with this mutation are more prone to staurosporine-induced cell death highlighting its cytoprotective functionality. In human postmortem brain tissue, synphilin-1, like a-synuclein is present in neurophil.

Biochem/physiol Actions
Synaptophysin (also known as the major synaptic vesicle protein p38) is a protein encoded by the SYP gene in humans. It is expressed in paneth cells, in goblet cells (in the mucus and cytoplasm) and in the neurons/synapses in the mucosa and submucosa. The gene plays a critical role in regulating neurotransmitter release and synaptic plasticity. It may be primarily associated with attention-deficit/hyperactivity disorder (ADHD). The gene is found to be involved in the susceptibility to schizophrenia. Abnormal expression of this gene in different brain regions may be linked to disorder in postmortem brain.

SYT4 (synaptotagmin 4) protein is thought to play a role in human brain functioning and neurological disorders. SYT4 null mice die shortly after birth. SYTL4 (Synaptotagmin-like protein 4) gene encodes a protein belonging to the synaptotagmin like protein family. These proteins consist of an N-terminal Rab27 binding domain and C-terminal tandem C2 domain. The C-terminal domain binds to Ca2+ and phospholipids trafficking the secretory vesicles to the plasma membrane. SYTL4 protein binds certain Rab GTPases and functions in intracellular membrane trafficking. It regulates exocytosis of dense-core granules by interacting with Rab27.

Biochem/physiol Actions
Transforming acidic coiled-coil-containing protein 3 is a protein encoded by the TACC3 gene in humans. It is referred as ERIC1 and ERIC-1. It is a member of the human transforming acidic coiled-coil (TACC) family of centrosomal adaptor proteins. It plays a critical role in microtubule nucleation at the centrosome. It is involved in the regulation of microtubule nucleation at the centrosome and functions in the stabilization of the γ-tubulin ring complex assembly. It plays an essential role in spindle assembly and centrosome integrity during mitosis as well as for cellular survival. It may act as a potential therapeutic target in cancer cells. It is aberrantly expressed in a variety of human cancers. It acts as a driver of tumorigenesis as well as an inducer of oncogenic EMT.

Biochem/physiol Actions
TAF7 (TATA-box binding protein associated factor 7) acts as a transcriptional repressor, where it binds to TAF1, and prevents its acetyl transferase activity. This activity is necessary for the basal transcription of MHC (major histocompatibility class) I. It acts as a regulator of check-point and prevents premature initiation of transcription until the preinitiation complex (PIC) is assembled completely. It interacts with multiple transcription factors such as, Sp1 (specificity protein), YY1 (Yin Yang 1), USF (upstream transcription factor 1), CTF (C-terminal fragment), adenovirus E1A, and HIV-1 (human immunodeficiency virus) Tat (trans-activator of transcription). It is also involved in the activation of thyroid hormone and vitamin D3 receptor in a ligand-independent fashion. It might also be involved in the processing of the 3′ end of mRNA.

TAGLN (transgelin) is an actin stress fiber binding protein. It is mainly involved in cell growth, differentiation, invasion, muscle fiber contractility and matrix remodeling. TAGLN is up-regulated in osteosarcoma cell lines, colorectal adenocarcinoma, prostate , hepatocellular , gastric, pancreatic and colon cancer. However, it is down-regulated in urinary bladder, renal cell carcinoma and colorectal cancer.

Target description
Transgelin (UniProt: Q01995; also known as 22 kDa actin-binding protein, Protein WS3-10, Smooth muscle protein 22-alpha, SM22-alpha) is encoded by the TAGLN (also known as SM22, WS3-10) gene (Gene ID:6876) in human. Transgelin is a shape change sensitive actin-binding protein of the calponin family that contains a calponin-homology domain (aa 24-137) and a C-terminal calponin-like module (aa 175-200) that is essential for actin binding. It is found in vascular and visceral smooth muscles of all adult vertebrates and is considered as an early marker of smooth muscle differentiation. Transgelin expression has also been reported in fibroblasts, spinal cord, adrenal gland, and in several immune cells. It is overexpressed in senescent human fibroblasts. It is shown to be transiently expressed in heart and skeletal muscle cells during early embryogenesis in mouse. Transgelin serves as an actin stress fiber-associated protein that stabilizes actin gels in vitro. It is reported to promote podosome formation and contributes to cell motility in response to injury or inflammation. Activation of TGF 1 signaling is shown to induce Transgelin expression, which enhances mesenchymal stem cells (MSC) differentiation and migration. Over-expression of Transgelin is shown to enhance osteoblast and adipocyte differentiation of MSC and these effects are abolished in the presence of TGF beta signaling inhibitors. Elevated levels of Transgelin are reported to promote tumor metastasis and are correlated with aggressive tumor behavior and poor prognosis in colorectal cancers. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Target description
Talin-1 (UniProt: Q9Y490) is encoded by the TLN1 (also known as KIAA1027, TLN) gene (Gene ID: 7094) in human. Talin-1 is a peripheral membrane protein that localized in adhesion complexes between cells and the extracellular matrix. It is shown to colocalize with layilin at the membrane ruffles and localized preferentially in focal adhesions than fibrillar adhesions. Talin-1 is involved in connections of major cytoskeletal structures to the plasma membrane. It binds to and activates integrins, directly connecting integrins to the actin cytoskeleton and recruiting numerous proteins, such as vinculin and focal adhesion kinase into integrin adhesion complexes. Talin-1 contains a globular N-terminal head region and a flexible rod domain, which can be dissociated by the action of calpain 2. The head region contains a FERM (protein 4.1, ezrin, radixin, moesin) domain that has binding sites for the cytoplasmic domains of -integrins, layilin, and F-actin. The head region is also known to bind PIPK1 90 and the focal adhesion kinase. The rod domain is known to have an additional integrin-binding site, two actin-binding sites, and several binding sites for vinculin. Talin-1 expression is upregulated in several human cancers and accelerates tumor cell migration and metastasis. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage.
