Biochem/physiol Actions
TBL1XR1 (transducin β-like 1X-related protein 1) regulates the change from gene repression to activation of nuclear receptors and proteins involved in transcriptional activation. It also functions as an E3-ligase. TBL1XR1 haploinsufficiency results in intellectual disability, without autistic behavior. Mutations in TBL1XR1 are linked with autism spectrum disorders. TBL1XR1 is involved in progression of breast cancer cells by transactivation of cyclin D1 and activation of the β-catenin signaling pathway.

Specificity

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected Tbr2 in NK-92 cell lysate.
Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Tbr2 in E16.5 mouse embryo tissue sections.

Target description
Eomesodermin homolog (UniProt: O95936; also known as T-box brain protein 2, T-brain-2, TBR-2) is encoded by the EOMES (also known as TBR2) gene (Gene ID: 8320) in human. T-box genes are a family of transcription factors have a highly conserved DNA binding domain named the T-domain. Tbr2 functions as a transcriptional activator that plays a crucial role during gastrulation, mesodermal specification, and limb patterning. It plays a role in brain development and is required for the specification and the proliferation of the intermediate progenitor cells and their progeny in the cerebral cortex. It is considered as the major intrinsic determinants of the intermediate neural progenitors. It is reported to be transiently expressed in cortical subventricular zone progenitor cells. Tbr2 is also involved in the differentiation of CD8+ T-cells during immune response regulating the expression of lytic effector genes. Its DNA-binding region (T-Box) is localized to amino acids 276-456. Conditional inactivation of Tbr2 during early brain development is known to cause microcephaly and severe behavior deficits. Four isoforms of Tbr2 have been described that are produced by alternative splicing. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Biochem/physiol Actions
TBX5 (T-box 5) plays a key role in the morphogenesis of vertebrate cardiac system. It controls cardiac growth and development by acing as an inducer of cardiac cell division arrest. Mutations in this gene result in cardiac abnormalities, such as septation defects and isomeris. Mutations also result in deformation of upper limbs and Holt-Oram syndrome (HOS), which is an autosomal dominant disorder. Mutations in this gene are also associated with arrhythmias and non-synodromic/synodromic congenital heart diseases. Loss of function mutation in this gene also results in familial dilated cardiomyopathy.

Target description
Transcription factor 7-like 2 (UniProt: Q9NQB0; also known as HMG box transcription factor 4, T-cell-specific transcription factor 4, T-cell factor 4, TCF-4, hTCF-4) is encoded by the TCF7L2 (also known as TCF4) gene (Gene ID: 6934) in human. The mammalian T-cell factor family consists of four members (TCF1 4) that contain a highly conserved high-mobility-group (HMG) domain that is responsible for their binding to DNA. TCF-4 is the most prominent member of this family that is highly expressed in small intestine with highest expression at the top of crypts and a gradient expression from crypt to villus. It is also detected in colon epithelium and colon cancer and in mammary gland carcinoma. However, its expression in fetal small intestine is barely detectable. HMG box of TCF-4 is localized to amino acids 350-418. TCF-4 participates in the Wnt signaling pathway and modulates Myc expression by binding to its promoter in a sequence-specific manner. It acts as a repressor in the absence of catenin b1, and as an activator in its presence. It is shown to activate transcription from promoters with several copies of the Tcf motif 5-CCTTTGATC-3 in the presence of catenin b1. Transducin-like enhancer of split 1,2,3, and 4 (TLE1-4) are reported to repress transactivation mediated by TCF-4 and catenin b1. TCF-4 undergoes phosphorylation at Thr 201 or Thr 212 by Nemo-like kinase (NLK), which inhibits its DNA-binding and prevents transcriptional activation of target genes of the canonical Wnt/b-catenin signaling pathway. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb® formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage.
