Specificity
Clone 2N10-H1 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects human GDP-L-fucose synthase. It targets an epitope within 20 amino acids from the C-terminal region.

Target description
GDP-L-fucose synthase (UniProt: Q13630; also known as EC:1.1.1.271, GDP-4-keto-6-deoxy-D-mannose-3,5-epimerase-4-reductase, Protein FX, Red cell NADP(H)-binding protein, Short-chain dehydrogenase/reductase family 4E member 1) is encoded by the GFUS (also known as SDR4E1, TSTA3) gene (Gene ID: 7264) in human. In mammals, fucosylated glycans play important roles in ABO blood group antigens, host-microbe interactions, and several human diseases, including rheumatoid arthritis, pancreatic cancer, and hepatocellular carcinoma. GDP-L-fucose synthase is a homodimeric protein of the NAD(P)-dependent epimerase/dehydratase family that is responsible for the last step of the major metabolic pathway resulting in GDP-L-fucose synthesis from GDP-D-mannose. It catalyzes a combined epimerase and NADPH-dependent reductase reaction, converting GDP-4-keto-6-D-deoxymannose to GDP-L-fucose. This reaction involves epimerization at both C-3 and C-5 followed by an NADPH-dependent reduction of the carbonyl at C-4. GDP-L-fucose synthase is also reported to act as the main autoantigen for a brain-infiltrating T cell clone. It is recognized by cerebrospinal fluid-infiltrating CD4+ T cells from HLA-DRB3*-positive subjects. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form
Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
