Quick Notes:- This kit contains the KAPA3G HotStart DNA Polymerase enzyme, enabling probe-based qPCR for both routine and challenging sample types.
- Initial denaturation of 3 min at 98°C is recommended to ensure complete denaturation of complex target DNA. A 5-min denaturation time may be required for some crude samples.
- For two-step cycling, use a 20-sec combined annealing/extension/data acquisition at 60°C as a first approach.
- A 10-sec annealing/extension/data acquisition time may be used with most assays, but this must be determined empirically.
- For crude samples, the amount of sample in the reaction may be reduced to improve performance, but this must be determined empirically.

Biochem/physiol Actions
The KAPA Taq EXtra PCR system is a blend of KAPA Taq DNA polymerase and a modified archaeal (Type B) DNA polymerase possessing proofreading capability. This two-enzyme system is designed specifically to support robust, long range, and sensitive PCR. Both enzymes possess 5→3 polymerase activity, but only KAPA Taq DNA polymerase possesses 5→3 exonuclease activity and only the Type B DNA polymerase possesses 3→5 exonuclease (proofreading) activity. Reaction products generated with KAPA Taq EXtra are polyA-tailed, and can be cloned into TA-cloning vectors.

NACRES: NA.54 General descriptionKAPA2G Fast Genotyping Mix (2X) is a ready-to-use master mix containing all components for fast polymerase chain reaction (PCR), except primers and template. The ReadyMix™ also contains two inert dyes, allowing for the analysis of PCR reaction products by agarose gel electrophoresis without the need to add a DNA loading solution. The KAPA Express Extract system contains a thermostable protease, designed for rapid sample lysis. The KAPA Express Extract, a novel thermostable protease and buffer system allows the extraction of PCR-ready DNA from mouse tissue in as little as 15 minutes, and KAPA2G Fast Genotyping Mix with dye, containing a DNA polymerase engineered via directed evolution for high processivity and extreme speed. Biochem/physiol Actions
DNA extracted with KAPA Express Extract is amplified with KAPA2G Fast Genotyping Mix with dye. DNA fragments generated with KAPA2G Fast Genotyping Mix are 3-dA-tailed and may be cloned into TA cloning vectors, or used for routine downstream analyses or applications, including restriction enzyme digestion, cloning and sequencing. Like wild-type Taq, KAPA2G Fast has 53 polymerase and 53 exonuclease activities, but no 35 exonuclease (proofreading) activity. The fidelity of KAPA2G Fast is like that of wild-type Taq; it has an error rate of approximately 1 error per 1.7 x 105 nucleotides incorporated. Lysis is performed in a thermocycler, heating block or water bath, after which the sample is centrifuged to recover the DNA containing supernatant. The supernatant is then diluted 10-fold before use as a template in PCR. Unlike existing protocols that rely on Proteinase K digestion, extractions using KAPA Express Extract are conveniently performed in a single tube, without the need for hazardous chemicals, or multiple wash steps. This greatly reduces the risk of sample loss and contamination. The process yields enough template for multiple assays and is easily scaled to handle samples in 96-well format. Each extraction yields enough template for at least 100 genotyping PCR reactions. Extracts (either diluted or undiluted) are stable at -15°C to -25°C for at least 6 months.

KAPA2G Robust DNA polymerase has 5→3 polymerase and 5→3 exonuclease activities, but no 3→5 exonuclease (proofreading) activity. The fidelity of KAPA2G Robust is similar to that of wild-type Taq; it has an error rate of approximately 1 error per 1.7 x 105 nucleotides incorporated. KAPA2G Robust is a highly processive DNA polymerase, capable of amplifying DNA fragments of up to 1 kb in size with a 15 sec/cycle extension time. The use of excessive extension times is likely to result in smearing and nonspecific amplification. For amplicons <1 kb with genomic DNA as a template, 15 sec per cycle should be sufficient for a high yield of the product. In the case of longer amplicons, the time may be increased to a maximum of 60 sec/kb, in 15-sec increments. PCR products generated by KAPA2G Robust are 3-dA-tailed and may be cloned into TA cloning vectors.

Addition of DMSO in the reaction at a final concentration of 1-4% may increase yield and improve reliability of the system with some complex PCR targets.Other NotesAccuTaq LA DNA polymerase is an optimized blend of Sigma′s high quality Taq DNA polymerase and a small amount of an additional polymerase that exhibits 3′→5′ exonuclease or proofreading activity. Principle
The WTA2 process involves two steps. In the first step, sample RNA is reverse transcribed with non-self-complementary primers composed of a quasi-random 3′ end and a universal 5′ end. During this process, displaced single strands serve as new templates for primer annealing and extension. The resultant cDNA library, comprised of random, overlapping 100 - 1000 base fragments flanked by universal end sequence. The Expand 20 kbPLUS PCR System is composed of a special enzyme mix containing thermostable Taq DNA polymerase and Tgo DNA polymerase, a thermostable DNA polymerase with proofreading activity. This powerful polymerase mixture and associated buffer system is designed to give a high yield of PCR product when fragments longer than 20 kb need to be amplified.
T/A cloning can be used for fragments produced by this kit.
To obtain long PCR products, first evaluate length and purity of the DNA template. Handling all genomic DNA templates with care. Genomic DNA template should be larger than 50 kb, as determined with agarose gel electrophoresis.
The Expand 20 kbPLUS PCR System contains human control DNA and human control -globin primers which allow the amplification of a 23 kb fragment.
These reagents may serve as a control reaction but can also be used to test the quality of human template DNA s and/or the respective primer pairs.
T/A cloning can be applied.
For a standard 50 µl PCR, we recommend using 5 U of the enzyme blend.
