Biochem/physiol Actions
DDAH2 (Dimethylarginine dimethylaminohydrolase 2) exists in two isoforms, DDAH1 and DDAH2. It is highly expressed in tissues expressing endothelial nitric oxide synthases (NOS). It controls the level of methylarginine. Inhibited activities of the protein can lead to increased concentration of (asymmetrical dimethylarginine) ADMA in endothelial cells. This increased ADMA further inhibits the NO-mediated endothelium dependent relaxation of blood vessels.

Biochem/physiol Actions
DDRGK1 (DDRGK domain containing 1) gene encodes a member of the DDRGK domain-containing protein family. The expression of this protein is induced by ER stress. It is found to interact with a protein complex containing UFL1 (UFM1-specific ligase 1), the putative tumor suppressor LZAP/C53 and UFM1 (ubiquitin fold modifier 1). UFL1 and LZAP/C53 are involved in the regulation of NF-κB pathway. DDRGK1 interacts with and stabilizes IκBα (inhibitor of NF-κB) and in turn functions in the regulation of transcriptional activity of NF-κB and the expression of its target genes. Loss of DDRGK1 activity is found to inhibit cell proliferation.

The gene DDX42 (DEAD-box helicase 42) encodes a component of 17S U2 snRNP splicing complex. It is found to associate with the Japanese encephalitis virus (JEV) NS4A (nonstructural protein 4A) protein. This interaction inhibits the stimulation of an innate immune response to JEV infection. The C-terminal tail of the protein interacts with ASPP2 (apoptosis-stimulating protein of p53 2) and regulates its apoptosis-stimulating properties.

The exact function of DDX47 (DEAD (Asp-Glu-Ala-Asp) box polypeptide 47) is not well understood, but it is thought that this protein plays a role in the biosynthesis of ribosomes. This protein interacts with the C-terminal of the E1E4 protein of human papillomavirus type 16 (HPV16), and might be involved in the replication of HPV16. This protein is also a binding partner of GABA(A) receptor-associated protein (GABARAP), which acts a chaperone for GABA (γ-aminobutyric aid)(A) receptor in the neurons of cortex. Together, GABARAP and DDX47 might be involved in apoptosis. DDX47 is involved in the processing of pre-rRNA, and is recruited to the pre-rRNA processing region by NOP132 (nucleolar protein).

Biochem/physiol Actions
DDX56 (DEAD, Asp-Glu-Ala-Asp, box helicase 56) is a putative ATP-dependent RNA helicase mainly involved in the necleolar activity. During ribosome synthesis, it helps to assemble 60S ribosomal subunit, and in unwinding of nucleic acid duplexes. Its DEAD-box motif is responsible for the intrinsic ATPase activity, which varies depending on the polynucleotide presence. DDX56 also participates in ribosome biogenesis. It is also associated with the virion assembly pathway. Its enzymatic action alters the ability to establish infection of West Nile virus (WNV) particles.
