Biochem/physiol Actions
EMP2 (epithelial membrane protein 2) is involved in blastocyst implantation, where it controls the surface expression and localization of integrin αvβ3. It is up-regulated in endometrioid ovarian tumors, which is associated with poor prognosis and survival. It is involved in controlling the correct trafficking of glycolipids and multiple proteins from post-Golgi endosomes to plasma membrane. It also regulates the proper trafficking of specific molecules to glycolipids-enriched lipid raft microdomains (GEMs). It interacts with and activates focal adhesion kinase (FAK) and the FAK/Src complex, which in turn facilitates collagen gel contraction. This gene is also down-regulated in nasopharyngeal carcinoma (NPC), especially in high grade cases. In NPC, it might be involved in increases tumor aggressiveness, and poor prognosis. It is highly up-regulated in triple negative breast cancer and invasive breast cancer. It might have potential as a therapeutic target for the same.

EMX1-lineage derived cells are responsible for two types of neuronal population in mature basal forebrain namely, a subclass of distinct amygdala excitatory neurons, and medium spiny neurons (MSNs) in the striatum. Target description
Endoglin (UniProt: P17813; also known as CD105) is encoded by the ENG (also known as END) gene (Gene ID: 2022) in human. Endoglin is a single-pass type I, disulfide-linked, membrane glycoprotein that is detected on placenta and umbilical vein endothelial cells. It can also form heterodimeric complex with the signaling receptors for TGFb. It is synthesized with a signal peptide (aa 1-25), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 26-586), a transmembrane domain (aa 587-611), and a cytoplasmic domain (aa 612-658). Endoglin also contains a zona pellucida (ZP) domain (aa 363-533) that mediates its dimerization. Endoglin is reported to play an important role in the regulation of angiogenesis. It is also required for normal structure and integrity of adult vasculature. It regulates the migration of vascular endothelial cells and regulates the change in shape of endothelial cells in response to blood flow thereby causing vascular remodeling and establishment of normal vascular morphology during angiogenesis. It has been also reported to play a role in the binding of endothelial cells to integrins and/or other RGD receptors. Endoglin also serves as a co-receptor for TGFb and is involved in the TGFb/BMP signaling cascade leading to the activation of SMAD transcription factors. Mutations in ENG gene have been linked to hereditary hemorrhagic telangiectasia that is characterized by recurrent epistaxis and gastro-intestinal hemorrhage. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.
Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Biochem/physiol Actions
ENDOU (Endonuclease, polyU-specific) is a serine protease glycoprotein belonging to the XendoU family. It is a placenta specific protein expressed in the syncytiotrophoblast and in various tumor tissues. In humans, it is involved in the generation of molecules with 2,3-cyclic phosphate ends. It cleaves at uridylates of single stranded RNA in presence of Mn2+ through its RNA binding capability. It has been reported that ENDOU may have biomarker characteristics for identifying the level of various cancers including ovarian adenocarcinoma.

Target description
Nitric oxide synthase, endothelial (UniProt: P29474; also known as EC:1.14.13.39, Constitutive NOS, cNOS, EC-NOS, Endothelial NOS, eNOS, NOS type III, NOSIII) is encoded by the NOS3 gene (Gene ID: 4846) in human. NOS is known to exist in three forms (a) a soluble, constitutively expressed enzyme found in high concentrations in the brain (nNOS or NOS-1), (b) a constitutively expressed endothelial membrane bound enzyme (eNOS, NOS-3), and (c) an inducible enzyme (iNOS or NOS-2) that is associated with the cytotoxic function of macrophages. They exhibit similarities in their structure and mechanism of action. Calmodulin is required for the activity of all three isoforms. All NOS are single polypeptides that differ in their rates of Nitric oxide ( NO), Ca2+- dependence, cytochrome c reduction, and NADPH utilization. For their catalytic activities NOS isoforms require three distinct domains: (a) a reductase domain, (b) a calmodulin-binding domain, and (c) an oxygenase domain. The reductase domain contains the FAD and FMN moieties. The oxygenase domain catalyzes the conversion of L-arginine to citrulline and NO. The eNOS is mainly expressed in vascular endothelial cells and cardiomyocytes. The NO produced by the eNOS in endothelial cells acts as a vasodilator and regulates blood flow and blood pressure. In cardiac cells, eNOS is expressed in response to shear stress, exercise, chronic hypoxia, and cardiac failure. In endothelial cells, eNOS is localized in specialized plasmalemmal pockets known as caveolae, which contain a scaffolding protein termed caveolin. Acylation of eNOS by either myristate or palmitate targets it to caveolae, which facilitates enzyme activation following receptor stimulation. In a resting cell, eNOS is inhibited by its interaction with caveolin. Following agonist activation, eNOS dissociates from caveolin and translocates to the cytosol. The eNOS is generally activated in response to an increase in intracellular Ca2+ following stimulation with receptor-dependent agonists such as acetylcholine. However, shear stress can also activate this enzyme, independently of Ca2+ level changes, via a tyrosine kinase-dependent mechanism involving its phosphorylation and redistribution. Low concentrations of NO in the endothelial cells prevent their apoptotic death and maintain their integrity during inflammation and atherosclerosis. Distorted production of NO in endothelial cells has been linked to hypertension, hypercholesterolemia, diabetes, and even cardiac failure. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Biochem/physiol Actions
APA (aminopeptidase A) that is encoded by ENPEP gene cleaves acidic residues, such as aspartate and glutamate, from the N- terminal of its substrates and its activity is modulated by calcium levels. It is one of the key enzymes in the central renin-angiotensin system, and regulates hypertension. It converts angiotensin II to angiotensin III by cleaving the N-terminal aspartate residue. gp160 plays a key role in the development of kidney, and therefore is also known as the human kidney differentiation antigen. APA activity has been shown to be decreased in various renal cell carcinomas as compared to normal renal cells. This might lead to an increase in the level of angiotensin II in renal carcinomas, hence contributing to angiogenesis. It has been suggested that the formation of Amyloid β peptides is facilitated by APA, where it truncates the full length Amyloid β at the N-terminal.

Epithelial cell adhesion molecule (EpCAM) might function as a hepatic stem/progenitor cell-specific marker. EpCAM is an oncogene activated by deliverance of its intracellular domain, which is involved in transducing signals into the cell nucleus by associating with elements of the wnt pathway. Since the epithelial cell adhesion molecule (EpCAM) is highly expressed in premalignant hepatic tissues and in a subset of hepatocellular carcinoma (HCC), it might act as an early biomarker of hepatocellular carcinoma (HCC). EpCAM gene mutation leads to a rare autosomal recessive diarrheal disorder in infants called congenital tufting enteropathy (CTE). Like other cell adhesion molecules, EpCAM plays a crucial role in cell-cell interaction and it is also associated with tight junction protein, like claudin-7. Experimental studies hypothesize that EpCAM is involved in normal intestinal and crypt villus axis development .

Epithelial cell adhesion molecule (UniProt: P16422; also known as Ep-CAM, Adenocarcinoma-associated antigen, Cell surface glycoprotein Trop-1, Epithelial cell surface antigen, Epithelial glycoprotein, EGP, Epithelial glycoprotein 314, EGP314, hEGP314, KS 1/4 antigen, KSA, Major gastrointestinal tumor-associated protein GA733-2, Tumor-associated calcium signal transducer 1, CD326) is encoded by the EPCAM (also known as GA733-2, M1S2, M4S1, MIC18, TACSTD1, TROP1) gene (Gene ID: 4072) in human. Ep-CAM is single-pass type I membrane monomeric protein that may act as a physical homophilic interaction molecule between intestinal epithelial cells and intraepithelial lymphocytes at the mucosal epithelium for providing immunological barrier as a first line of defense against mucosal infection. It plays a role in embryonic stem cells proliferation and differentiation. It is selectively expressed by undifferentiated cells and its levels decline rapidly upon embryonic stem cell differentiation. Its expression is observed in almost all epithelial cell membranes, but not on mesodermal or neural cell membranes. It is also reported to be expressed on the surface of adenocarcinoma. On carcinoma tissue it is found in a hyperglycosylated state. Glycosylation at asparagine 198 is shown to be essential for its stability. EpCAM is synthesized with a signal peptide (aa 1-23), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 24-265), a short transmembrane domain (aa 266-288), and the cytoplasmic domain (aa 289-314). Mutations in EPCAM gene have been linked to Hereditary non-polyposis colorectal cancer 8 that is characterized by a familial predisposition to early-onset colorectal carcinoma and extra-colonic tumors of the gastrointestinal, urological and female reproductive tracts. This ZooMAb recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

EPX (eosinophil peroxidase) is a heme-containing enzyme. It is also a pro-angiogenic protein and stimulates endothelial cells to control vessel development. Levels of EPX in sputum is connected with eosinophil cell counts in the sputum. Single nucleotide polymorphism in this gene is associated with inflammatory bowel disease. This association is dependent on the age and gender of the person. Humans with ulcerative colitis show high intraluminal release of EPX. EPX is also linked with various cancers. Absence or low activity of EPX causes hereditary eosinophil peroxidase deficiency.

Target description
Receptor tyrosine-protein kinase erbB-3 (UniProt: P21860; also known as EC:2.7.10.1, Proto-oncogene-like protein c-ErbB-3, Tyrosine kinase-type cell surface receptor HER3) is encoded by the ERBB3 (also known as HER3) gene (Gene ID: 2065) in human. ErbB3 is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-19), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 20-643), a transmembrane domain (aa 644-664), and a cytoplasmic domain (aa 665-1342). Its extracellular domain is formed of two ligand-binding regions (L1 and L2) and two cysteine-rich regions (CR1 and CR2). In the absence of a ligand, the extracellular domain stayed in a locked conformation and is stabilized by interactions between CR1 and CR2 regions that prevents its dimerization. Its tyrosine kinase domain is localized to amino acids 709-966 and shares homology with EGFR and ErbB2 kinase domain. However, the tyrosine kinase activity is impaired due to modified residues and its activation requires the formation of heterodimers with other EGFR family receptors. Known ligands for ErbB3 include neuregulins (NRG) and NRG1 and NRG2 appear to be the most active. Downstream signaling pathways activated include MAPK and PI3 Kinase pathways. ErbB3 has been implicated in cell division and survival and its expression is essential for cell proliferation in several tumor models and transformation of normal tissues. It has also been implicated in acquired resistance to EGFR-tyrosine kinase inhibitors. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb® formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Receptor tyrosine-protein kinase erbB-2 (UniProt: P04626; also known as EC:2.7.10.1, Metastatic lymph node gene 19 protein, MLN 19, Proto-oncogene Neu, Proto-oncogene c-ErbB-2, Tyrosine kinase-type cell surface receptor HER2, p185erbB2, CD340) is encoded by the ERBB2 (also known as HER2, MLN19, NEU, NGL) gene (Gene ID: 2064) in human. ErbB2 is a single-pass type I membrane glycoprotein that is synthesized with a signal peptide (aa 1-22), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 23-652), a transmembrane domain (aa 653-675), and a cytoplasmic domain (aa 676-1255). Its tyrosine kinase domain is localized to amino acids 720-987. It can also be translocated to the nucleus and its nuclear localization signal resides in amino acids 676-689. It has intrinsic protein tyrosine kinase activity and it mediates the lateral signal transduction of all ErbB receptors. It is involved in in the regulation of a variety of cellular functions, including cell growth, differentiation, and apoptosis. It also regulates outgrowth and stabilization of peripheral microtubules. In the nucleus it is involved in transcriptional regulation and associates with the 5-TCAAATTC-3 sequence in the PTGS2/COX-2 promoter and activates its transcription. It is also implicated in transcriptional activation of p21. ERBB2 gene is reported to be overexpressed in about 30% of all human breast carcinomas. ErbB2 is activated by dimerization and autophosphorylation. Ligand-binding is shown to increase phosphorylation on tyrosine residues.
