HK1 (Hexokinase 1) encodes a hexokinase protein consisting of a porin-binding domain at the N-terminal end. It is majorly involved in the glucose metabolic pathway. It is expressed in the photoreceptor inner section, outer plexiform layer, inner nuclear layer, inner plexiform layer, and ganglion cell layer of the human retina. It provides the energy for maintaining the metabolic activity of retina. In the mitochondria, HK1 functions as a pro-survival factor to inhibit TNF-induced apoptotic signals. A missense mutation in hexokinase 1 (HK1) causes an autosomal dominant disorder, retinitis pigmentosa. It has been reported that porin-binding domain of HK1 actively participates in the mitochondrial integrity.

HLA class II histocompatibility antigen, DM α chain (HLA-DMA) is majorly involved in antigen presentation to cluster of differentiation-4 (CD4) T-cells. It catalyzes the release of class II-associated invariant chain-derived peptides (CLIP) from class II histocompatibility molecules which are newly synthesized. It has a peptide exchange activity and functions as a chaperone to peptide-free class II major histocompatibility (MHC) molecules. HLA-DMA binds to class II molecules, like HLA-DR for catalysis and the inhibitor HLA-DO.

The gene HLA-DOB (major histocompatibility complex, class II, DO β) encodes the β chain of HLA-DO, a non-classical class II heterodimer that functions as a modulator of antigen presentation. HLA-DO associates with HLA-DM and inhibits the peptide loading function. HLA-DPB1 (major histocompatibility complex, class II, DP β 1) recognizes and presents antigens to T-helper cells. Polymorphisms in this gene are linked to the differences and level of response to rubella vaccination. Variants of this gene are associated with systemic sclerosis, where DPB1*03:01 and *13:01 alleles are up-regulated. HLA-DQA1 is a component of human leukocyte antigen (HLA) molecule. The allele contains two chains: a DQα and a DQβ chain. It is a class II gene, expressed at the membrane of antigen presenting cells (APC) in heterodimeric form. It is mainly involved in the immune response to foreign antigens by presenting specific antigenic peptides to T cells. The gene is associated with a sleep disorder narcolepsy with cataplexy. An association of the gene with celiac disease and type 1 diabetes has been reported in studies.

HLTF (helicase-like transcription factor) interacts and binds with DNA to control transcription. It utilizes the energy of ATP hydrolysis for chromatin remodeling, which is basic to multiple cellular processes. It plays a role in post-replication DNA repair, where it functions as E3 ubiquitin ligase and uniquitinates proliferating cell nuclear antigen (PCNA). This protein, along with translesion synthesis polymerases, is recruited to chromatin by the tumor suppressor protein BRCA1 (breast cancer 1, early onset). In stalled damaged DNA replication, this protein repairs the gaps in the replication fork. HLTF is hypermethylated and inactivated in multiple cancers such as, esophageal, gastric, colorectal and uterine cancer. Its expression correlates with the progression of thyroid neoplasia. Thus, it has potential as a marker to differentiate benign from malignant thyroid tumors. In cervical cancer, it increases the DNA repair capacity, and thus, confers resistance to radiotherapy.

Target description
High mobility group protein B1 (UniProt: P09429; also known as High mobility group protein 1, HMG-1) is encoded by the HMGB1 (also known as HMG1) gene (Gene ID: 3146) in human. HMG-1 is a ubiquitously distributed, multifunctional redox sensitive protein with various roles in different cellular compartments. In basal state predominantly nuclear. It shuttles between the cytoplasm and the nucleus. Translocates from the nucleus to the cytoplasm upon autophagy stimulation. In the nucleus, as one of the major chromatin-associated non-histone proteins, it acts as a DNA chaperone involved in replication, transcription, chromatin remodeling, V(D)J recombination, DNA repair, and genome stability. It promotes host inflammatory response to sterile and infectious signals and is involved in the coordination and integration of innate and adaptive immune responses. Its HMG box 1 and 2 regions are localized to amino acids 9-79 and 95-163, respectively. HMG box 2 is reported to mediate proinflammatory cytokine-stimulating activity and binding to TLR4. In the cytoplasm it dissociates the BECN1:BCL2 complex via competitive interaction with BECN1 leading to autophagy activation. It is also involved in oxidative stress-mediated autophagy. In its cytoplasmic role it also functions as sensor and/or chaperone for immunogenic nucleic acids implicating the activation of TLR9-mediated immune responses. Biochem/physiol Actions
HMGCL (3-hydroxymethyl-3-methylglutaryl-CoA lyase) is a homodimeric mitochondrial enzyme. It is a key modulator of ketogenesis and l-leucine catabolism. During ketogenesis, it catalyzes the cation-dependent cleavage reaction to form acetyl-CoA and acetoacetate. This cleavage reaction generates ketone bodies to support the energy requirements. Its activity is highly dependent on the divalent cation such as Mg2+, Mn2+ and stimulated by reducing agents (DTT). Deficiency of HMGCL causes a rare autosomal recessive genetic disorder, 3-Hydroxy-3-methylglutaric aciduria.

Target description
Hepatocyte nuclear factor 1 (UniProt: P35680; also known as HNF-1-beta, HNF-1B, Homeoprotein LFB3, Transcription factor 2, TCF-2, Variant hepatic nuclear factor 1, vHNF1) is encoded by the HNF1B (also known as TCF2) gene (Gene ID: 6928) in human. HNF-1 is a transcription factor of the HNF1 homeobox family that binds to the inverted palindrome 5-GTTAATNATTAAC-3. It binds to DNA as a dimer. It controls endoderm development and is shown to be crucial for pancreatic and renal development. It is also involved in the hepatobiliary specification of hepatoblasts to cholangiocytes during liver development and is strongly expressed throughout adult biliary epithelium. Over-expression of HNF-1 is associated with pathological subtype of primary tumor and its expression in hepatocellular carcinoma is associated with the change of phenotype on recurrence. Mutations in HNF1B are known to cause Renal cysts and diabetes syndrome (RCAD), an autosomal dominant disorder comprising non-diabetic disease resulting from abnormal renal development. HNF1B knockout mice are shown to suffer from severe neonatal cholestasis and jaundice due to the abnormally developed gall bladder and dysfunctional intrahepatic bile ducts. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage.
