Specificity
Clone 1H12 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects human Leukocyte immunoglobulin-like receptor subfamily B member 4 (LILRB4). It targets an epitope within 19 amino acids from the extracellular domain.

Anti-LILRB4, clone 1H12 ZooMAb®, Cat. No. ZRB1113, is a rabbit recombinant monoclonal antibody that specifically targets LILRB4 and is tested for use in Affinity Binding Assay, Flow Cytometry, Immunocytochemistry, and Western Blotting.
Quality Control TestingEvaluated by Western Blotting in MDA-MB-435 lysate.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected LILRB4 in MDA-MB-435 cell lysate.
Tested ApplicationsImmunocytochemistry Analysis: A 1:100 dilution from a representative lot detected LILRB4 in NK-92 cells.
Affinity Binding Assay: A representative lot of this antibody bound LILRB4 peptide with a KD of 2.5 x 10-7 in an affinity binding assay.

Target description
Leukocyte immunoglobulin-like receptor subfamily B member 4 (UniProt: Q8NHJ6; also known as CD85 antigen-like family member K, Immunoglobulin-like transcript 3, ILT-3, Leukocyte immunoglobulin-like receptor 5, LIR-5, Monocyte inhibitory receptor HM18, CD85k, LILRB4) is encoded by the LILRB4 (Gene ID: ILT3, LIR5) gene (Gene ID: 11006) in human. LILRB4 is a single-pass type I membrane protein that serves as a receptor for class I MHC antigens and recognizes a broad spectrum of HLA-A, HLA-B, HLA-C and HLA-G alleles. It is involved in the downregulation of the immune response and the development of tolerance, such as towards transplants. It is reported to inhibit receptor-mediated phosphorylation of cellular proteins and mobilization of intracellular calcium ions. LILRB4 expression has been reported in monocytes, macrophages, dendritic cells, lung, natural killer cells and B-cells and is induced upon contact with CD8+ CD28- alloantigen-specific T suppressor (Ts) cells. LILRB4 is also a tumor-associated antigen that is highly expressed on monocytic AML cells where its expression is more specific and can be used to differentiate monocytic AML (M5) from AML M1-M3. It is synthesized with a signal peptide (aa 1-21), which is subsequently cleaved off to generate the mature form that contains an extracellular domain (aa 22-259), a transmembrane domain (aa 260-280), and a cytoplasmic domain (aa 181-448). It contains three copies of immunoreceptor tyrosine-based inhibitor motif (ITIM) that are involved in modulation of cellular responses and the phosphorylated ITIM motif can bind the SH2 domain of several SH2-containing phosphatases. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form
Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
