Biochem/physiol Actions
Pyruvate kinase (PK) is a key enzyme in the glycolytic pathway. Knockdown of the M2 isoform in human cancer cell lines and its replacement by the M1 isoform has been shown to lead to reversal of the Warburg effect, and reduced ability to form tumors in mouse xenografts. Phosphorylation of the M2 isoform at Tyr105 inhibits its activity and is common in human cancers, suggesting that Tyr105 is a critical metabolic switch in cancer cells that promotes tumorigenesis.

Biochem/physiol Actions
The gene PKP2 (plakophilin 2) encodes a desmosomal protein that belongs to the plakophilin family of proteins that are involved in desmosome formation and function. Desmosomal proteins participate in cell signaling. In mouse, Pkp2 is expressed mainly in the cardiac muscle and plays an important role in the heart development. Any mutation in this gene causes defects of heart morphogenesis and junctional architecture with embryonic lethality at midgestation. Mutations in this gene are also associated with arrhythmogenic right ventricular cardiomyopathy.

PLA2G3 (group 3 secretory phospholipase A2) is a secretory phospholipase. It is involved in the release of cellular arachidonate (AA). PLA2G3 is reported as a negative regulator of ciliogenesis. Absence of PLA2G3 results in cells showing centrosome-concentrated recycling endosomes. In human neuronal system, it participates in xanthine/xanthine oxidase (X/XOD)-mediated apoptotic cell death. Polymorphism in PLA2G3 is linked with Alzheimer′s disease (AD). The expression of PLA2G3 is deregulated in various malignant tissues.

PLA2G6 (phospholipase A2, group VI) gene encodes a phospholipase that catalyzes the release of fatty acids from phospholipids. The protein may function in phospholipid remodeling, nitric oxide-induced or vasopressin-induced arachidonic acid release. It is also involved in leukotriene and prostaglandin synthesis. It may have a role in fas mediated apoptosis and in regulating transmembrane ion flux in glucose-stimulated B-cells. It is also involved in cardiolipin (CL) deacylation. Mutations in PLA2G6 are associated with infantile neuroaxonal dystrophy, neurodegeneration with brain iron accumulation type II and autosomal recessive early-onset dystonia-parkinsonism. Loss of PLA2G6 gene activity causes lipid peroxidation, mitochondrial dysfunction and thereby mitochondrial membrane abnormalities. It is also involved in type 1 diabetes, by participating in endoplasmic reticulum stress-induced β-cell apoptosis.

Target description
Secretory phospholipase A2 receptor (UniProt: also known as PLA2-R, PLA2R, 180 kDa secretory phospholipase A2 receptor, C-type lectin domain family 13-member C, M-type receptor) is encoded by the PLA2R1 (also known as CLEC13C) gene (Gene ID: 22925) in human. PLA2R is a single-pass type I membrane protein that serves as a receptor for secretory phospholipase A2 (sPLA2). It is highly expressed in kidney and is also present in lung macrophages, pancreas, amnion, and placenta. PLA2R is a receptor for phospholipase sPLA2-IB/PLA2G1B, but not sPLA2-IIA/PLA2G2A. Target description
Phospholipase D2 (UniProt: O14939; also known as EC:3.1.4.4, PLD 2, hPLD2, Choline phosphatase 2, PLD1C, Phosphatidylcholine-hydrolyzing phospholipase D2) is encoded by the PLD2 gene (Gene ID: 5338) in human. PLD2 is a peripheral membrane protein that displays ubiquitous expression and plays a role in cell membrane lipid reorganization and serves as a key cell signaling protein. It is activated by a wide variety of G-protein coupled receptors and receptor tyrosine kinases in response to a wide variety of agonists. It hydrolyzes the membrane phospholipid phosphatidylcholine to choline and the lipid messenger phosphatidic acid (PA). PA can act as a central lipid second messenger for various signaling pathways and participates in many physiological processes, including cell proliferation, migration, and cytoskeletal organization. PA is also involved in the activation of RasGTPase by recruiting its exchange factor Sos or Rac1to the plasma membrane. PLD2 is reported to positively regulate proliferation of CD8+ T cells that play a critical role in cancer cell elimination and suppression of tumor progression. PLD2 is reported to degrade HIF-1 and thereby reduces VEGF secretion by endothelial cells that results in reduction of tumor neovascularization and growth. PLD2 has PX and PH domains at its regulatory Nterminus that are putatively responsible for its GEF activity. It also has two conserved HKD domains are responsible for the lipase activity and conversion of PC into PA. Its catalytic region is localized within amino acids 441-788. Biochem/physiol Actions
Phospholipase D3 (PLD3) influences amyloid-β precursor protein (APP) processing. It is highly expressed in those regions of the brain which are vulnerable to Alzheimer′s disease pathology, including hippocampus and cortex. Overexpression of PLD3 leads to the decrease in intracellular APP and extracellular Aβ42 and Aβ40 (the isoforms of the amyloid-β peptide). On the other hand, knockdown of PLD3 leads to an increase in extracellular Aβ42 and Aβ40.

PLEKHM1 (pleckstrin homology and RUN domain containing M1) is involved in the end stages of endocytic and autophagy pathways. Here, it interacts with RAB7 (member of RAS oncogene family), the HOPS (homotypic fusion and vacuole protein sorting) complex and ATG8 (autophagy-related protein 8) proteins. It plays an important role in autophagosome-lysosome fusion and endosome and autophagosome maturation. PLEKHM1 also participates in osteoclast function and bone resorption. Plasminogen receptor KT (PLGRKT) plays a role in plasminogen-dependent monocyte and macrophage migration, invasion and recruitment in the inflammatory response. Biochem/physiol Actions
PLOD3 (procollagen-lysine, 2-oxoglutarate 5-dioxygenase 3) encodes a membrane-bound homodimeric enzyme that catalyzes the hydroxylation of lysyl residues and O-glycosylation of hydroxylysyl residues in collagen-like peptides. The hydroxylysyl groups of these hydroxylysines act as sites of attachment for carbohydrate units and stabilize intermolecular collagen cross-links. The enzyme is localized to the cisternae of the rough endoplasmic reticulum. The gene is mapped to human chromosome 7q36. Deficiency of lysyl hydroxylase activity is observed in some patients with Ehlers-Danlos syndrome type VIB.

Biochem/physiol Actions
Proteolipid protein (PLP) is involved in membrane adhesion, compaction of myelin and synthesis of myelin intra-period line. It is also involved in the maturation of oligodendrocytes. PLP plays a role in wrapping, maintenance and survival of axons. Mutations in the proteolipid protein 1 (PLP1) gene is associated with Pelizaeus-Merzbacher disease indicated by nystagmus, spasticity, microcephaly, ataxia and intellectual disability. PLP1 gene plays a role in Xq22.2 microdeletion and microduplication syndromes. Duplication of this gene results in the overexpression of PLP, which leads to accumulation in the cytoplasm myelinating oligodendrocytes and defects in myelin.
