NACRES: NA.46 General descriptionImmunoglobulin G (IgG) belongs to the immunoglobulin family and is a widely expressed serum antibody. It consists of a gamma (γ) heavy chain in the constant (C) region. The monomeric 150 kDa structure of IgG constitutes two identical heavy chains and two identical light chains with molecular weight of 50 kDa and 25 kDa, respectively. The primary structure of this antibody also contains disulfide bonds involved in linking the two heavy chains, linking the heavy and light chains and resides inside the chains. IgG is further subdivided into four classes namely, IgG1, IgG2, IgG3, and IgG4 with different heavy chains, named γ1, γ2, γ3, and γ4, respectively. Immunoglobulin M (IgM) exists as a pentamer in the serum and has a high molecular weight. Monomeric IgM has a molecular weight of 190 kDa. The pentameric IgM consists of ten μ-heavy chains, ten light chains and a single J-chain which is linked by disulphide bonds. The μ-chain consists of five domains- VH, Cμ1, Cμ2, Cμ3 and Cμ4. Immunoglobulin A (IgA) is found as two subclasses, that is, IgA1 and IgA2. IgA1 has an extended hinge because of the insertion of a duplicated stretch of amino acids, whereas IgA2 lacks this. IgA exists as a monomer or dimer in the secreted form. Dimeric IgA has a molecular weight of 390 kDa. This antibody can form multimers and its polymerization is essential for transport across epithelia. IgA is crucial as the first line of defence.

Biochem/physiol Actions
IgG4 is the least abundant immunoglobulin present in the human serum. IgG4 is implicated in a unique process called fab-arm exchange (FAE) and limits the formation of immune complex. It has anti-inflammatory properties. IgG4 functions as a therapeutic antibody. It provides protection against allergy, prevents mast cell degranulation and has a harmful role in malignant melanoma. IgG4 may be associated with autoimmune diseases such as, autoimmune pancreatitis (AIP) type 1 and 2, Mikulicz syndrome, Kuttner′s tumor, Riedel′s thyroiditis and tubulointerstitial nephritis.
IgG antibody subtype is the most abundant of serum immunoglobulins of the immune system. It is secreted by B cells and is found in blood and extracellular fluids and provides protection from infections caused by bacteria, fungi and viruses. Maternal IgG is transferred to fetus through the placenta that is vital for immune defense of the neonate against infections. The coupling of biotin to monoclonal Anti-Human IgG4 antibody allows for the binding of various labels such as avidin or streptavidin.

IgM is an initial immunoglobulin produced in the developing fetus as a result of the immune response during the second trimester of pregnancy. It is then substituted by IgG. Therefore, the diagnosis of specific antibodies of the IgM class is useful in detecting chronic infections. IgM is essential for the fetal and neonatal host defense system. IgM cannot cross the placenta. Hence, the presence of IgM might indicate an intrauterine viral infection. IgM antibodies are responsible for the agglutination of red blood cells in mismatched blood transfusions. The level of IgM may vary with the status of disease or infection.

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide as preservative
Storage and Stability
For continuous use, store at 2-8 °C for up to one month. For extended storage, the solution may be frozen in working aliquots at –20 °C. Repeated freezing and thawing, or storage in "frost-free" freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use.
